In the course of cleaning and painting yesterday, I noticed that the nylon (is it nylon?) thru hull on the port side of the transom (which I think is the bilge pump discharge, have to check) is loose. The question is, if I unscrew the outside part enough to put caulk underneath will the job be done? OR: Is it necessary to unscrew completely to fill the whole underside of the part with caulk? Should I use polysulfide for this job? If I can just loosen, will it tighten back up by just screwing from the outside or is it a two man job for somebody to be in the depths of the locker holding the inside part so it doesn't turn? Also, don't think I ever saw a special tool for screwing this part down with a slot on either side of the hole. Is there a special tool for this? Thank you all as always

If this is a typical hard plastic thru-hull, there's likely a hose clamped to the inboard end and a nut. If so, you'll have to remove the hose before turning the exposed head of the thru-hull. Or you might get away with just loosening the nut from the inside, pushing the thru-hull out a fraction of an inch, and carefully working some sealant under the flange of the head from outside of the boat, then retightening the nut.
If the thru-hull is near or below the waterline, I'd be a bit more thorough about it, by completely removing the thru-hull, carefully inspecting it for cracks, cleaning all old sealant from the parts and hole, then reassembling with fresh sealant and two new hose clamps.
As I recall, polysulfide is specifically <b>NOT</b> reccommended for use with plastics. Silicone would be my choice here. Others might also suggest polyurethane.
FWIW, I use 4200 above the waterline and 5200 below.
Thr through hull you are describing should be for the manual bilge pump unless someone has rearranged the plumbing. I just replaced mine. I had a friend hold it still with two large screwdrivers while I tightened the locking nut from the rear berth. It had been removed while I repaired and applied new gel coat to the rensom after upgrading the ladder to a swim platform/ladder combination. The original holes were filled and new holes were drilled. Well, I cracked the thru hull from overtightening. The plastic was brittle from age. After a quick trip to WM I replaced it with a new one.
You asked about removing it or just squeezing some sealant under it. If it were me I would remove it and clean all the old sealant. It seems to me that you might have voids doing it the other way. I would pull it, scrape of the old sealant, apply a liberal amount of 4200, then replace it. You can wipe off the excess 4200 after tightening the lock nut.
My boat has an inboard so I guess I'll have get into the locker to tighten it. Do I need to let the lifeseal setup for a week before I tighten it down. Here I thought I was finally down to the polishing, spiffing up stage, oh well.
